Two-dimensional concentration maps through spectral estimation and spectral unmixing

Abstract

We developed a spectral imaging microscope that relies only on emission signals with no additional optics added to the microscope. Thus, it can resolve relative concentration in samples with dimensions below the diffraction limit and there is no need to see the actual structure of a sample to determine its composition as long as it emits light. From im­age colour we estimate the spectral emission of the sample. From the estimated fluorescence emission of multiple overlapping microscopic species, we demonstrate the microscope's spectral unmixing perform­ance on light emitting diodes (LEDs), co-localized multi-color single-stained fluorescent microspheres, and multi-stained chromosomes and generate their two dimensional concentration maps.
